April 3, 2026Pancreatic ductal adenocarcinoma (PDAC) is characterized by a dense stroma and dysfunctional vasculature, limiting drug delivery and immune infiltration, while melanoma, though less fibrotic, remains highly metastatic and treatment-resistant. To overcome these barriers, we employed a combinatorial approach repurposing the antihistamine ketotifen with ultrasound-mediated sonopermeation to normalize the Tumor Microenvironment (TME) and enhance chemotherapy efficacy in murine models of PDAC and melanoma. The triple therapy suppressed tumor growth in both models, with pronounced effects in PDAC, where it reduced stiffness, restored vascular function and improved perfusion, alleviated hypoxia, and enhanced T-cell infiltration, compared to untreated controls.
